| I. Background | |
| CD47 是一种广泛表达于多种细胞表面的跨膜糖蛋白基因，属于免疫球蛋白超家族成 员。其编码的蛋白质在细胞表面发挥重要功能，主要顺利获得与信号调节蛋白α（SIRPα）结合， 传递“别吃我”信号，抑制巨噬细胞等免疫细胞对自身细胞的吞噬，从而在免疫逃逸和自 身耐受中起关键作用。在肿瘤微环境中，癌细胞常高表达 CD47，借此逃避免疫监视，促 进肿瘤生长与转移。此外，CD47 还参与细胞迁移、增殖及凋亡等过程，并与血小板活化、 神经发育等功能相关。研究表明，CD47 在多种血液系统恶性肿瘤及实体瘤中高表达，与 不良预后相关，使其成为肿瘤免疫治疗的潜在靶点。现在，针对 CD47 的阻断抗体或融合 蛋白等疗法正在临床研究中，旨在恢复巨噬细胞对肿瘤细胞的吞噬作用，为癌症治疗给予 新策略。 | |
